But yes it's based on how they identify. If I were born a man and become a woman who preferred men I would be straight. Biological sex means nothing.

You are either male or female at birth (Hemaphrodites aside) and this is unrelated to whether you want to mash genitalia with males/females/both/nothing. Although both are influenced by biology and environment, they don't determine each other.

I would imagine so, but that makes it a bit more of a relative point. You would be considered a lesbian if your identity was considered to be female, but if it was considered to be male regardless then you would not be.

tbh I dunno cuz i had a girlfriend for a while, and now I'm here. But honestly, even though my grandfather is gay, I think it's still a matter of nurture over nature, possibly the absence of a father figure for boys. Girls I dunno about, but who honestly cares about lesbians?
