No, I killed Gehrman then got the ending where you replace him. I reloaded the save and found I had only consumed 2/4 of the cords.

I always thought the Queen/Locust were from those labs but they never followed it up after GoW 2, which more than annoyed me. They also changed the queen from looking like a Locust, to just a human in some costume...
It was explained in some books I think, at least the lambet were.
